利用染料結合的方法來定量檢測釋放到組織培養液或從生物材料中提取的彈性蛋白。
 
應用范圍:樣本可來源于細胞培養基和含細胞外基質成分的組織提取物(如彈性軟骨、主動脈、皮膚等),可檢測可溶性彈性蛋白原和不可溶性彈性蛋白(抽提后以可溶性彈性蛋白多肽的形式檢測)。
 
樣本要求:樣本體積5-100ul,包含5-75ug的彈性蛋白
 
實驗操作步驟

彈性蛋白的分離:用彈性蛋白沉淀劑處理樣本,使彈性蛋白沉淀下來。
彈性蛋白與染料結合:裝有沉淀的彈性蛋白管中,加染料和反應液,反應60min以上
彈性蛋白-染料復合物獲得:去掉未結合的染料
復合物染料的釋放:添加染料釋放劑,使彈性蛋白-染料復合物中的染料釋放出來
釋放出的染料檢測:繪制標準曲線,513nm或405nm處定量檢測
